Overview
I am an Associate Professor in Biochemistry with over twenty years of academic and research experience at the University of Insubria and the University of Pavia. My primary research focus lies in extracellular matrix (ECM) biology, specifically investigating the metabolism and regulation of glycosaminoglycans (GAGs), such as hyaluronan and heparan sulphate, and their critical roles in vascular remodelling, inflammation, and cancer microenvironments. My scientific track record includes 71 indexed publications with an h-index of 35 (Scopus), alongside a proven history of securing national and international research funding, including European Commission and joint bilateral grants.
Beyond my research activities, I possess extensive experience in university governance and academic leadership, currently serving as the President of the Biomedical Laboratory Techniques Degree Course and having contributed as an institutional board member for University Quality Assurance (PQA) and Research Grant Distribution (FAR). On a national level, I was involved in research evaluation, serving as a GEV 5 member for the Italian Research Assessment (VQR 2020–2024) under ANVUR. My international standing is reflected in my role as an expert grant evaluator for the Horizon Europe program and the National Science Centre (NCN) in Poland, as well as my active contributions to international scientific societies and editorial boards within the field of matrix biology.

Research Interests
The main research focus is the metabolism of glycosaminoglycans (GAGs), specifically hyaluronan (HA) and heparan sulphate (HS) polysaccharides:
• Synthesis and polymerisation mechanisms of HA mediated by the hyaluronan synthase enzyme family (HAS1-3) on the plasma membrane, and the differential regulation of HAS2 and HAS3 in adult cells and tissues.
• Synthesis of HS chains within the ER and Golgi on the core proteins of specific proteoglycans (PGs), with a focus on the regulation of the polymerising and enzymatic complex known as the GAGosome (composed of EXT polymerising enzymes, sulfatases, and epimerases).
• Metabolic alterations of HA and HSPGs (e.g., Syndecans) induced by cytokines during inflammation or cancer, affecting both HAS expression and HS sulfation patterns.
• The role of alterations in the endothelial glycocalyx (EC) and smooth muscle cell (SMC) extracellular matrix (ECM) induced by inflammatory cytokines and oxidised LDL. Studies show these particles increase HA synthesis (consistent with in vivo data on intima thickening during atherosclerosis), leading to increased monocyte recruitment/activation and altered vascular permeability.
• The effects of LDL/cholesterol (or their oxidised forms) on the EC glycocalyx, particularly regarding the role of HAS3 (which appears to be affected in its expression but not its enzymatic activity).
• Investigating the role of PCSK9 and its potential interactions and systemic sequestration by membrane-bound HS.

The Glycocalyx–Cytoskeleton Axis: An Integrated In Vitro Model of Endothelial–SMC Crosstalk and Vascular Remodelling in Hypertension
The underlying structural causes of vascular injury often remain untreated. Consequently, millions of patients continue to develop progressive and frequently irreversible vessel damage. Early hypertension induces inflammation, reduced nitric oxide bioavailability, and increased vascular permeability, disrupting the protective endothelial glycocalyx–vascular muscle axis. This initiates a feedback loop that promotes vessel wall stiffening, extracellular matrix remodelling, and long-term tissue damage.
This project investigates how the endothelial glycocalyx, extracellular matrix, and stress-related chaperone proteins contribute to hypertensive vascular damage. To overcome the limitations of animal models, the project involves developing a human cell-based platform spanning isolated cells, microfluidic systems, and 3D culture.
In this context, this line of research focuses specifically on extracellular matrix biology, with particular attention to the synthesis and organisation of hyaluronan (HA), collagen, and other matrix components directly involved in vascular stiffening. Leading expertise in this field allows for the detailed characterisation of extracellular matrix remodelling as the project progresses from cellular studies to human validation.
By analysing endothelial responses and using flow-based systems within microfluidic devices, cellular alignment, permeability, and the structural integrity of the glycocalyx and endothelial surface are assessed. The gathered findings will guide the development of advanced three-dimensional vascular scaffolds designed to faithfully reproduce native vessel architecture and mechanical properties.
The clinical validation of the model includes the involvement of patients across different blood pressure phenotypes. By studying alterations on the endothelial surface exposed to patient-derived serum, the objective is to identify early biomarkers by correlating circulating molecular signatures with disease stage, while evaluating the efficacy of targeted molecules in preventing vascular damage.
The project aims to establish a predictive three-dimensional human vascular model for safer, faster, and more ethical in vitro screening of drugs and biomarkers in accordance with the 3Rs principles. Ultimately, it aims to shift the therapeutic focus from blood pressure control to the prevention of structural vascular deterioration, enabling earlier diagnosis and improved cardiovascular outcomes.
